The grant aims to provide a proof-of-principal for a novel strategy to identify misprocessed and aberrant proteins in Tau diseases using the proteogenomic approach. It encourages collaborative efforts to create advanced single-cell or single-cell type proteogenomic platforms. The research objectives include creating a comprehensive database of misprocessed and aberrant proteins in selected mouse models of human Tau diseases, cross-validating the presence of misprocessed and aberrant proteins in human AD/ADRD brains, identifying new molecular pathways and novel misprocessed protein-protein interaction networks, defining novel mechanisms through which misprocessed and aberrant proteins influence neurodegeneration, and identifying disease-specific therapeutic targets. Applications that only provide a global view of gene expression without any subcellular, cell-type, and brain regional specificity will be considered non-responsive.

Eligibility Requirements
Eligible applicants include higher education institutions, nonprofits, for-profit organizations, state and local governments, federally recognized Indian/Native American tribal governments, federal agencies, U.S. territories or possessions, public housing authorities/Indian housing authorities, Native American tribal organizations (other than federally recognized tribal governments), faith-based or community-based organizations, regional organizations, and foreign organizations. Foreign components of U.S. organizations are also eligible to apply.

Period of Performance
The period of performance for the grant is up to 5 years.

Grant Value
The National Institute on Aging (NIA) intends to commit $4,000,000 in FY 2024 to fund 4-6 awards. Application budgets may not exceed $500,000 in direct costs per year.
