Project Title Purchase order for gene synthesis and plasmid preparation from GenScript USA Statement of Need/Purpose and Objectives Our lab studies the folding and dynamics of fold-switching proteins, in particular the use of deep learning AI systems such as AlphaFold to successfully predict unique structures from sequences with high homology to known proteins. This leads to a research loop of using deep learning systems to select candidate sequences that are predicted to fold into suitable structures, expressing and purifying these proteins in bacterial systems, using structural techniques such as nuclear magnetic resonance (NMR) and mass spectrometry to determine how well these proteins match their predicted structures, and to use the experimental results to refine the next round of in silico studies to further optimize the sequences. In order to maintain our pace of research, we require the rapid synthesis of DNA sequences for the expression of new protein constructs. The specific sequences and thus the exact DNA constructs cannot be determined in advance and instead will be optimized based on the results of ongoing experiments. Having access to an open purchase order will ensure a minimum of turnaround time when new sequences are required.
