Background

This contract aims to procure sequencing services necessary for generating time series data to monitor ecosystem changes, particularly in relation to West Coast Ocean Acidification and related investigations.

Work Details
The contractor will provide library preparation and sequencing services for eDNA samples collected from various marine environments. Specific tasks include:

1. Library preparation involving indexing/barcoding, normalization, and quality control of PCR amplicon products derived from seawater eDNA filters.
2. Sequencing of a total of 6,912 DNA libraries across six distinct metabarcoding loci, accommodating up to 1,152 samples.
3. Ensuring that the sequencing services meet specific requirements for time series generation, including maintaining molecular biology best practices to prevent contamination.
4. Conducting a two-step PCR amplification and library preparation protocol with unique dual indexing for each library.
5. Quality checking the indexed amplicon library size and concentration prior to sequencing, implementing appropriate size selection steps as necessary.
6. Coordinating with PMEL scientists throughout the project to ensure high-quality data production.

Period of Performance
The period of performance for this contract is from September 30, 2026, to September 29, 2027.

Place of Performance
The work will be performed at the contractor's facility capable of handling molecular biology tasks related to eDNA analysis.
