Background
The National Institutes of Health (NIH) National Heart Lung and Blood Institute is seeking to purchase the NanoAssemblr Ignite equipment to generate lipid nanoparticles (LNPs) for in vivo gene therapy. The purpose of this acquisition is to develop an HSC-targeted gene delivery system with lentiviral vectors (LVs) and LNPs, allowing for the development of in vivo HSC-targeted gene therapy. This system aims to enable a single-injection cure of sickle cell disease (SCD) on a walk-in basis worldwide, including in low-resource countries. The project is being conducted by the NHLBI Cellular and Molecular Therapeutics Branch, along with other laboratories and the NHLBI non-human primate program.

Work Details
The NanoAssemblr Ignite equipment is intended to constantly generate uniform LNPs for gene delivery. It will be used for scale-up from in vitro experiments to animal experiments, including humanized mice and rhesus macaques, as well as translation to humans. The instrument is designed for drug developers using their own materials to formulate nanoparticles, with formulation volumes ranging from 1 mL to 15 mL.

The Ignite platform of instruments simplifies the transition into clinical development and manufacturing, resulting in the automated synthesis of genomic medicines with minimal setup and training.
