Description

Posted: March 5, 2026, 9:17 p.m. EST
The Defense Advanced Research Projects Agency (DARPA) is soliciting innovative approaches to controllably eliminate invasive species from defined areas, primarily focused on New World Screwworm (NWS) and Brown Tree Snake (BTS)(Guam), using genetic drive technologies (GDT). GUARDIAN is composed of three Technical Areas (TAs). The first Option TA is focused on BTS eradication. Proposers are expected to complete modeling and laboratory testing of genetic elements in the 18-month Phase 1 and contained (laboratory or greenhouse) testing during the 18-month Phase 2. Option TA1 will emphasize: genetic precision to ensure off-target effects and GDT resistance is minimized; GDT control by designing counter measures/GDT immunization to add additional safeguards; and fieldability by ensuring laboratory and contained testing meets regulatory agency requirements to proceed to field trials and that the GDT meets requirements needed for impactful population reduction goals. Option TA2 seeks to develop innovative cell cultures methods that accelerate the study of gene drives in eukaryotic organisms and should contain elements that support Option TA1. The third Option TA focuses on population modeling of gene drives, particularly that of NWS and BTS.
The purpose of Amendment 1 to DARPA-PS-26-17, the GUARDIAN (Genetic Utilization for Advanced Regulation and Defense of Indigenous and Native species) Program Solicitation (PS), is to clarify the submission instructions and proposal abstract content.

Background
The Defense Advanced Research Projects Agency (DARPA) is seeking innovative solutions to control and eliminate invasive species, specifically the New World Screwworm (NWS) and Brown Tree Snake (BTS) in Guam, utilizing genetic drive technologies (GDT).

The GUARDIAN program consists of three Technical Areas (TAs), with a focus on developing effective methods for species eradication while ensuring minimal off-target effects and regulatory compliance for field trials.

Work Details
The contract involves multiple phases across three TAs:
- TA1 focuses on BTS eradication through transgenics creation over two 18-month phases. Key tasks include:
- Phase 1: Development of genetic constructs, modeling, and laboratory testing.
- Milestones include presentations on construct design, delivery methods, and mid-phase demonstrations.

- TA2 aims to innovate cell culture methods for studying gene drives in eukaryotic organisms, supporting TA1's objectives.

- TA3 emphasizes population modeling of gene drives for NWS and BTS. Each phase has specific milestones that must be met to ensure progress and funding continuation.

Period of Performance
The GUARDIAN program spans a total of 36 months for TAs 1 and 3, consisting of an initial 18-month Phase 1 followed by an additional 18-month Phase 2.

TA2 operates solely within an 18-month Phase 1 timeframe.

Place of Performance
The contract work will primarily take place in laboratory or greenhouse settings as specified for contained testing.
