Description

The Defense Advanced Research Projects Agency (DARPA) is soliciting innovative concepts to design a protein complex that can be expressed in vivo (i.e., in a living cell) and responds to optical signals to synthesize a corresponding DNA or RNA sequence. This technology should enable massless transfer of genetic information to cells expressing the protein complex via a template-free mechanism that incorporates nucleotide bases in response to specific optical signals (e.g., different wavelengths of light). The GO program is focused on two main Research Objectives (ROs): (1) achieving the core capability of synthesizing DNA/RNA directly in living cells using optical signals, and (2) developing error mitigation methods that ensure high-fidelity nucleic acid synthesis. The acquisition process for GO will proceed through a two-stage process, starting with (1) written abstracts which will inform invitations to brief (2) an Oral Proposal Package (OPP) at DARPA.
Background
The Defense Advanced Research Projects Agency (DARPA) is seeking innovative concepts for the Generative Optogenetics (GO) program. This program aims to develop a protein complex that can be expressed in living cells and respond to optical signals for the synthesis of DNA or RNA sequences. This technology is intended to facilitate the massless transfer of genetic information using a template-free mechanism that reacts to specific optical signals.

The program focuses on two main research objectives: synthesizing nucleic acids directly in living cells using light signals and developing methods to ensure high-fidelity nucleic acid synthesis.

Work Details
The Performer will be responsible for the overall technical and program management, including technical planning and execution of the Program. Key tasks include:
1. Achieving the core capability of synthesizing DNA/RNA directly in living cells using optical signals.
2. Developing error mitigation methods for high-fidelity nucleic acid synthesis.
3. Continuous collaboration with DARPA, ensuring access to program results and rights in patents and data.
4. Documenting accomplishments of each milestone through reports as specified in Attachment 1.
5. Submitting a Data Management Plan within 30 days after award, detailing data sharing and preservation strategies.
6. Disclosing inventions related to the project within specified timelines and filing patent applications as necessary.

Period of Performance
The Program commences upon the effective date of the agreement and continues for a specified number of months, with provisions for extension based on research opportunities.

Place of Performance
The geographic location(s) for contract performance are not explicitly provided.
