Project Title Characterization of RfaH evolution and folding dynamics: pairing in vitro and in situ methods Statement of Need/Purpose and Objectives Open PO so that I can continue to order plasmids as I discover and design new RfaH fold-switchers and probe the interface of the fold-switching region for critical contacts necessary for fold-switching. This will include ordering gene synthesis based on DNA or protein sequences I provide that they can either send directly to me or insert into plasmids for protein expression in either HEK293 cells for FLIM FRET assays or in E. coli for protein purification for biophysical characterization studies. Genscript also already has the pDREAM vector as I have sent it to them during prior subcloning experiments. This reduces the cost as they have the plasmid on hand and I do not need to send it to them again. The purpose of this work is to determine how fold switching evolved in the NusG/Spt5 family of proteins and it is partially funding by mt PRAT fellowship.
