The purpose of the amendment is to extend the due date for proposals to February 14, 2025 at 5PM ET.

In addition, the amendment clarifies a discrepancy between the Cost Proposal Narrative and Cost Proposal Spreadsheet bundle attachments. Specifically, the amendment clarifies that any material and equipment item exceeding $5,000 requires back-up documentation (e.g., copy of catalog price lists, past purchase order or invoice, or vendor quotes prior to purchase).

The purpose of the amendment is provide a correction to the Bundle of Attachments. Specifically:

1. ARPA-H ADMIN & NATIONAL POLICY (Other Transaction) is updated to add paragraph 11. Synthetic Nucleic Acids Or Benchtop Nucleic Acid Synthesis Equipment.

2. ARPA-H TECH & MANAGEMENT is updated to correct the page limit to thirty-five (35) pages. In addition, the language regarding team members, subawardees, subcontractors, vendors, etc. has been updated to better reflect the program structure.

No changes were made to the other Bundle of Attachment documents.

Background
The Advanced Research Projects Agency for Health (ARPA-H) is initiating the Computational ADME Tox AnaLYsis for Safer Therapeutics (CATALYST) Program through this Innovative Solutions Opening (ISO).

The program aims to enhance drug development processes by leveraging advanced computational methods and innovative tools to improve the safety and efficacy of therapeutics.

ARPA-H anticipates multiple other transaction awards to support this initiative.

Work Details
The CATALYST Program consists of three main technical areas (TAs):

- TA1 focuses on developing a robust data repository for AI/ML applications, integrating diverse biomedical data types, and creating deep-learning models.
- TA2 aims to develop innovative living systems tools for model development, enhancing data collection and analysis through in vitro and ex vivo models.
- TA3 seeks to create advanced in silico human physiology models that provide comprehensive representations of human biology.

Each TA requires close coordination with others, detailed milestone planning, and regular reporting on technical and financial status to the ARPA-H Program Manager Team. Proposals must address potential obstacles, provide strategies for overcoming them, and include a detailed schedule or timeline for achieving project goals.

Period of Performance
The overall CATALYST timeline is structured as a 54-month effort consisting of two phases: Phase I lasting 36 months focused on developing predictive in silico platforms and Phase II lasting 18 months aimed at regulatory filings using digital-only data or hybrid models.
